Amyloid discordance analysis in cognitively normal monozygotic twins demonstrates that the memory domain is affected first in preclinical AD

چکیده

Abstract Background The earliest cognitive changes in Alzheimer’s disease (AD), even before amyloid‐beta (Aβ) is abnormal, remain largely unclear. We recently observed a cohort of cognitively normal older monozygotic twins that 15% twin‐pairs were discordant for Aβ status (Fig. 1). In this cohort, we investigated early by comparing decline over 4 years among (one twin and co‐twin abnormal Aβ), with both (concordant Aβ‐), Aβ+). expected the rate to depend on baseline least concordant Aβ‐ increasing from Aβ‐, Aβ+ twins. Method From EMIF‐AD PreclinAD study selected cognition at who had [ 18 F]flutemetamol PET scan one follow‐up available (n=188; 90 twin‐pairs) (Table defined group using visual read dynamic images. tested whether concordance was associated performance time two repeated measures memory, attention, executive function (EF) language domains linear mixed models adjusted age, sex, education genetic relatedness. Result At showed impaired all ( Fig. 2). Discordant lower memory scores compared (discordant Aβ+, p=0.02; p=0.04). Over time, found improved (p=0.002), did not change (p=0.15), while declined p=0.03; p=0.07). No significant other domains. Conclusion Our findings provide further support AD are domain subtle may already present amyloid pathology can be detected scan.

Discordance for ankylosing spondylitis in monozygotic twins.

Three monozygotic twin pairs, each over the age of 45 years, with ankylosing spondylitis are described. In two pairs there is discordance for this disease and in the third pair there is a marked difference in the severity of this disease. These findings provide evidence that environmental factors are necessary not only for the initial development of ankylosing spondylitis, but possibly also for...

Discordance for skeletal and cardiac defect in monozygotic twins.

A case of monozygotic male twins discordant for skeletal and cardiac defect is reported. One twin had the hemifacial microsomia type of the oculo-auriculo-vertebral dysplasia. The cotwin had no asymmetry of the face and normal ears, but preaxial polydactyly and ventricular and auricular septal defects. The cotwins were concordant for craniostenosis with a ridge metopic suture. Karyotypes were n...

Discordance for childhood coeliac disease in monozygotic twins.

Discordance for childhood coeliac disease in monozygotic twin boys is reported. The diagnosis of coeliac disease in the second-born twin was established by demonstration of a flat small intestinal mucosa on biopsy and a clinical response to a gluten-free diet. The first-born twin had a normal small intestinal mucosa. Phenotypic, genotypic, and dermatoglyphic observations as well as gross observ...

Nasal Septum and External Nasal Deformity Similarities in Monozygotic Twins and Paranasal Computed Tomography Analysis

Since identical twins occur as a result of the division of one egg, they have the same genetic structure. Therefore, their phenotypes and sex are also always the same. However, due to the effect of environmental factors, some of the characteristics of twins living in different regions appear to develop differently. In our case of 17-year-old maternal twins, we emphasized that nasal pathologies ...
